چکیده

Stress is the body’s reaction to any change that requires adaptive responses. In various organisms, stress a seizure-related comorbidity. Despite exposure stressors eliciting aversive behaviors in zebrafish, there are no data showing whether potentiates epileptic seizures this species. Here, we investigated previous an intense acute stressor positively modulates susceptibility pentylenetetrazole (PTZ)-challenged zebrafish. The conspecific alarm substance (CAS) was used elicit responses (3.5 mL/L for 5 min), observed by increased bottom dwelling and erratic movements. Then, fish were immediately exposed 7.5 mM PTZ 10 min induce seizure-like behaviors. seizure intensity, number of clonic-like (score 4), as well facilitated occurrence score 4 episodes decreasing latency which reached 4. Moreover, with heightened anxiety showed PTZ, since positive correlations between anxiety- found. Overall, CAS also whole-body cortisol levels our novel findings show prominent response PTZ-induced previously stressed reinforce growing utility zebrafish models assess comorbidities aiming elucidate how can affect vertebrates.

Arsenic and Oxidative Stress in Pentylenetetrazole-induced Seizures in Mice

Background and Objective: Chronic arsenic toxicity is a widespread problem; the role of brain oxidative stress has been suggested in the genesis of epilepsy and in the post-seizure neuronal death. However, studies investigating the effects of arsenic on seizure and related mechanisms are limited. The purpose of this study was to examine the effect of prolonged exposure to sodium arsenite on oxi...

Closed-loop neural stimulation for pentylenetetrazole-induced seizures in zebrafish

Neural stimulation can reduce the frequency of seizures in persons with epilepsy, but rates of seizure-free outcome are low. Vagus nerve stimulation prevents seizures by continuously activating noradrenergic projections from the brainstem to the cortex. Cortical norepinephrine then increases GABAergic transmission and increases seizure threshold. Another approach, responsive nervous stimulation...

Seizures Induced by Pentylenetetrazole in the Adult Zebrafish: A Detailed Behavioral Characterization

Pentylenetetrazole (PTZ) is a common convulsant agent used in animal models to investigate the mechanisms of seizures. Although adult zebrafish have been recently used to study epileptic seizures, a thorough characterization of the PTZ-induced seizures in this animal model is missing. The goal of this study was to perform a detailed temporal behavior profile characterization of PTZ-induced seiz...

Embryonic Exposure to Domoic Acid Increases the Susceptibility of Zebrafish Larvae to the Chemical Convulsant Pentylenetetrazole

BACKGROUND Domoic acid (DA) is a neurotoxin produced by diatoms of the genus Pseudo-nitzschia that targets the limbic system to induce tonic-clonic seizures and memory impairment. In utero DA exposure of mice leads to a reduction in seizure threshold to subsequent DA exposures in mid-postnatal life, and similar studies have shown neurotoxic effects in rats that were delayed until adolescence. ...

Valerenic acid and Valeriana officinalis extracts delay onset of Pentylenetetrazole (PTZ)-Induced seizures in adult Danio rerio (Zebrafish)

BACKGROUND Anticonvulsant properties have been attributed to extracts of the herbal medicine Valeriana officinalis. Our aims were to examine the anticonvulsant properties of valerenic acid and valerian extracts and to determine whether valerian preparations interact with the activity of other anti-epileptic drugs (phenytoin or clonazepam). To achieve these goals, we validated the adult zebrafis...
